目的阐明雄激素与前列腺癌中雄激素应答基因PTTG1(pituitary tumor transforming gene1)表达的关系。方法选择基因芯片筛选的大鼠前列腺雄激素应答基因PTTG1,应用Northern Blot检测其在去势大鼠补充雄激素Mib(Mibolerone)前后在前列腺组织中的表达。应用免疫组化检测PTTG1在人前列腺癌组织中的表达,进一步以Northern Blot检测雄激素依赖性前列腺癌细胞系LNCaP及雄激素非依赖性前列腺癌细胞系PC3和DU145中PTTG1的表达情况。结果PTTG1在去势7d的大鼠前列腺组织中没有表达,而补充Mib2d后即有显著的表达;PTTG1主要表达于人前列腺癌上皮细胞;0.1nmol/L Mib作用48h可显著上调LNCaP细胞PTTG1的表达;与LNCaP细胞比较,PC3和DU145中PTTG1的基础表达水平更高。结论雄激素可显著上调大鼠前列腺组织及人前列腺癌细胞中PTTG1的表达,提示PTTG1可能在人前列腺癌的发生、发展中发挥重要的作用。
Objective To elucidate the relationship between androgen and the expression of androgen receptor gene PTTG1 (pituitary tumor transforming gene1) in prostate cancer. Methods Rat prostate androgen responsive gene PTTG1 was screened by cDNA microarray. Northern Blot was used to detect the expression of PTTG1 in prostate tissue before and after administration of androgen Mib (Mibolerone) in ovariectomized rats. The expression of PTTG1 in human prostate cancer tissues was detected by immunohistochemistry and the expression of PTTG1 in androgen-dependent prostate cancer cell line LNCaP and androgen-independent prostate cancer cell lines PC3 and DU145 were further detected by Northern Blot. Results PTTG1 was not expressed in the prostatic tissue of rats after castration for 7 days, but was significantly increased after supplementation with Mib2d. PTTG1 was mainly expressed in human prostate cancer epithelial cells. Mice treated with 0.1 nmol / L Mib for 48 h significantly up-regulated PTTG1 expression in LNCaP cells ; The basal expression level of PTTG1 in PC3 and DU145 was higher than that in LNCaP cells. Conclusion Androgen can significantly up-regulate the expression of PTTG1 in rat prostate tissue and human prostate cancer cells, suggesting that PTTG1 may play an important role in the development and progression of human prostate cancer.
