目的:观察心肌梗死小鼠静脉移植成体心脏干细胞后,细胞在小鼠体内各器官的分布情况。明确心梗后静脉移植成体心脏干细胞在小鼠体内的分布和归巢情况。方法:分离培养小鼠心脏成体干细胞,采用流式细胞仪鉴定细胞,通过亲脂性染料CM-DiI标记细胞后行小鼠急性心肌梗死模型建立和细胞移植,分别在细胞移植后7、14、8天取小鼠心脏、肝脏、脑、脊髓、肺脏,行冰冻切片,在荧光显微镜下观察移植细胞在各组织器官存活和分布情况。结果:成体心脏干细胞分离培养后呈贴壁生长,流式细胞仪检测显示细胞纯度>80%。CM-DiI标记后荧光显微镜下观察可见标记的细胞胞浆胞核均被染成呈明亮的红色。心肌梗死后经静脉移植成体心脏干细胞,细胞在各组织中分布呈变化过程,7天时,在肺脏和肝脏分布较多,至14天和28天时,肺脏和肝脏分布减少,心脏分布逐渐增多,表现出向心脏的“归巢”现象,而脑和脊髓在28天的观察时间内分布较少。结论:采用CM-Di I标记心脏成体干细胞,操作简单,标记效果好,可用于短期的细胞体内追踪。小鼠心肌梗死后行经静脉成体心脏干细胞移植,28天后细胞在心脏的分布逐渐增多,表现出向心脏的“归巢”现象。
OBJECTIVE: To observe the distribution of cells in various organs of mice after intravenous transplantation of adult cardiac stem cells in myocardial infarction mice. The distribution and homing of adult cardiac stem cells in mice after vein grafting were confirmed. Methods: Adult mouse cardiac adult stem cells were isolated and cultured. Flow cytometry was used to identify the cells. The cells were labeled with lipofectamine CM-DiI, then established acute myocardial infarction model and cell transplantation. The heart, liver, brain, spinal cord and lung of the mice were taken from day and frozen sections were taken. The survival and distribution of transplanted cells in various tissues and organs were observed under fluorescence microscope. Results: Adult cardiac stem cells isolated and cultured after adherent growth, flow cytometry showed that the purity of cells> 80%. Under the fluorescence microscope, the nuclei of labeled cytoplasm of cells labeled with CM-DiI were bright red. After myocardial infarction, adult cardiac stem cells were transplanted intravenously. The distribution of cells in each tissue showed a process of change. At 7 days, there were more lung and liver distribution. At 14 days and 28 days, the distribution of lung and liver decreased and the distribution of heart increased gradually. Going to the heart’s “homing” phenomenon, while the brain and spinal cord were less distributed over 28 days of observation. Conclusion: CM-Di I labeled cardiac adult stem cells, simple operation, labeling effect is good, can be used for short-term in vivo cell tracking. After transcatheter arterial adult stem cell transplantation in mice after myocardial infarction, the distribution of the cells in the heart gradually increased after 28 days, showing a “homing” phenomenon to the heart.
