目的探讨血小板胶(APG)-胶原(Co)复合膜对大鼠牙周组织缺损的修复作用。方法将Co分别裁成5 mm×2 mm及7 mm×4 mm大小,抽取大鼠全血10 ml制作富血小板血浆(PRP),再将5 mm×2 mm Co浸润于PRP中制备APG-Co复合膜。将16只成年雄性Wistar大鼠随机平均分为实验组和对照组,分别制作大鼠下颌牙周骨缺损模型(缺损大小为5 mm×2 mm,磨除牙根面牙骨质)。实验组置APG-Co复合膜于缺损内,并于缺损表面覆盖7 mm×4 mm的Co。对照组仅用7 mm×4 mm Co覆盖缺损表面。术后4、6周每组分别处死4只,HE染色观察牙周组织缺损修复效果,Image-Pro Plus对术后6周HE染色结果进行分析,计算出新生牙周骨面积,应用SPSS 13.0软件对新生牙周骨面积进行统计学分析。结果实验组新生牙槽骨和牙骨质样组织的生成量和成熟度均优于对照组,术后6周实验组新生牙周骨面积显著高于对照组[(56 533.04±2 673.61)vs(23 006.05±2 198.10)μm2,P<0.05]。结论 APG-Co复合膜有显著的促进牙周骨组织缺损修复的作用。
Objective To investigate the repair effect of APG-Co on the periodontal defects in rats. Methods Co (Co) were cut into 5 mm × 2 mm and 7 mm × 4 mm respectively, 10 ml of rat whole blood was extracted to prepare platelet rich plasma (PRP), then 5 mm × 2 mm Co was infiltrated into PRP to prepare APG-Co Composite film. Sixteen adult male Wistar rats were randomly divided into experimental group and control group. Rat models of periodontal bone defect (defect size 5 mm × 2 mm, root deformity cementum) were made respectively. Experimental group APG-Co composite film in the defect, and the defect surface covered 7 mm × 4 mm Co. The control group covered the defect surface with only 7 mm × 4 mm Co. At 4 and 6 weeks after operation, 4 rabbits were sacrificed in each group. HE staining was used to observe the effect of repair of periodontal defects. Image analysis was performed 6 weeks after operation by Image-Pro Plus to calculate the bone area of newly diagnosed periodontal ligament. Results The formation and maturation of nascent alveolar bone and cementum in the experimental group were better than those in the control group, and the bone area in the experimental group at 6 weeks after operation was significantly higher than that in the control group (56 533.04 ± 2673.61 vs (23 006.05 ± 2 198.10) μm 2, P <0.05]. Conclusion APG-Co composite membrane has a significant role in promoting the repair of periodontal bone defects.
