目的建立快速检测转基因生菜中白藜芦醇和白藜芦醇苷的高效液相色谱法。方法采用Agilent-ZORBAX SB-C185μm4.6×250mm色谱柱,流动相为乙腈-水(45+55),流速1.0ml/min,波长303nm。结果线性关系良好,白藜芦醇r=0.9999,回收率86.0%~97.8%,变异系数6.6%,最低检出量0.0113μg;白藜芦醇苷r=0.9991,回收率81.6%~95.6%,变异系数7.0%,最低检出量0.0159μg。结论以高效液相色谱检测转基因生菜中白藜芦醇和白藜芦醇苷,操作方便、快速、准确度高。
OBJECTIVE To establish a rapid HPLC method for the determination of resveratrol and polydatin in transgenic lettuce. Methods Agilent-ZORBAX SB-C185 4.6 × 250mm column was used. The mobile phase consisted of acetonitrile-water (45 + 55), flow rate 1.0ml / min and wavelength 303nm. The results showed good linearity. Resveratrol r = 0.9999, recovery 86.0% -97.8%, coefficient of variation 6.6%, minimum detectable amount 0.0113μg; polydatin r = 0.9991, recoveries 81.6% -95.6% Coefficient of variation of 7.0%, the minimum detectable amount of 0.0159μg. Conclusion High performance liquid chromatography (HPLC) detection of resveratrol and polydatin in transgenic lettuce is convenient, rapid and accurate.
