肿节风水提物诱导白血病CEM细胞凋亡的机制

目的探讨肿节风水提物诱导CEM细胞凋亡作用的机制。方法用浓度不同的肿节风水提物对CEM细胞发生作用24、48、72h后,采用逆转录PCR技术检测凋亡相关基因bcl-2、bax和p53的表达,研究肿节风诱导白血病CEM细胞凋亡的机制。结果肿节风水提物能降低CEM细胞bcl-2基因的表达,并随药物浓度的升高、作用时间的延长,bcl-2基因的表达逐渐下降,作用48、72h后,与对照组比较有显著差异(P<0.05);同时能增强bax和p53基因表达,且随用药浓度的加大和作用时间的延长呈正比关系,与对照组比较有显著差异(P<0.05)。结论肿节风水提物诱导白血病CEM细胞凋亡的机制与下调bcl-2基因和上调bax基因的表达,使bcl-2/bax的比值下降,同时还与上调p53基因的表达有关。 Objective To investigate the mechanism of Fengjie feng shui inducing apoptosis of CEM cells. Methods CEM cells were treated with different concentrations of Aegilops taimen extract for 24, 48 and 72 hours. The expression of bcl-2, bax and p53 were detected by reverse transcription polymerase chain reaction (RT-PCR) Mechanism of apoptosis. Results Edema extract can decrease the expression of bcl-2 gene in CEM cells. With the increase of drug concentration and the prolongation of action time, the expression of bcl-2 gene gradually decreased. After 48 and 72 hours, compared with the control group (P <0.05). At the same time, the expression of bax and p53 gene was enhanced, and the expression of bax and p53 was increased with the increase of drug concentration and the prolongation of action time, which was significantly different from the control group (P <0.05). Conclusion The mechanism of Euthytiotrophomonastipuncture inducing apoptosis in leukemic CEM cells is related to down-regulating the expression of bcl-2 gene and up-regulating bax gene, decreasing the ratio of bcl-2 / bax, and upregulating the expression of p53 gene.
