目的:观察红芪多糖对实验性脾虚大鼠胃黏膜组织中COX-2表达的影响。方法:100%大黄水煎液连续42天灌胃制备脾虚证大鼠模型,造模后将大鼠随机分为6组,其中空白对照组、模型对照组正常饲养,药物组(高、中剂量组)用红芪多糖灌胃30天,对照组(高、中剂量组)以黄芪多糖灌胃30天进行干预;用免疫组化法检测大鼠胃黏膜组织中环氧合酶-2(cycloxygenase-2,COX-2)的表达率。结果:模型对照组COX-2阳性表达率与空白组比较显著升高(P<0.01),红芪多糖高剂量组,黄芪多糖高剂量组COX-2阳性表达率与模型组比较显著降低(P<0.01),红芪多糖中剂量组COX-2阳性表达率与模型组比较明显降低(P<0.05)。结论:红芪多糖能够影响脾虚大鼠胃黏膜中病理因子COX-2的表达,能提高黏膜修复能力,从而防止脾虚证胃黏膜发生进一步病变。
Objective: To observe the effect of Hedysari Polysaccharides on the expression of COX-2 in gastric mucosa of experimental rats with spleen deficiency. Methods: The rat model of spleen deficiency syndrome was prepared by intragastric administration of 100% rhubarb decoction for 42 days. After the model was established, the rats were randomly divided into 6 groups, including blank control group and model control group, Group) were treated with Radix Hedysari for 30 days. The control group (high dose group and middle dose group) were treated with Astragalus polysaccharide for 30 days. The expression of cyclooxygenase-2 in gastric mucosa of rats was detected by immunohistochemistry -2, COX-2) expression rate. Results: The positive rate of COX-2 in the model control group was significantly higher than that in the blank group (P <0.01). The positive rate of COX-2 in high dose Astragalus polysaccharide group and high dose Astragalus polysaccharide group was significantly lower than that in the model group <0.01). The positive rate of COX-2 in middle dose of Radix Hedysari was significantly lower than that in model group (P <0.05). Conclusion: Hedysari polysaccharide can affect the expression of COX-2, a pathological factor in the gastric mucosa of spleen-deficient rats, and can improve mucosal repair ability and prevent gastric mucosa from further pathological changes.
