目的:鉴定开心散60%乙醇提取物(KXS-60%E)及其血中移行成分中的远志皂苷类成分,分析大鼠体内远志皂苷类成分的代谢产物。方法:应用UPLC-TOF-MS技术,分析TOF-MS/MS给出的二级离子信息,结合液质联用工作站Mass Lynx v4.1分析软件及现有的文献数据库,鉴定KXS-60%E中远志皂苷类成分、灌服KXS-60%E后血清中的远志皂苷类成分及其代谢产物。结果:鉴定了KXS-60%E中14个远志皂苷类成分,主要为三萜型皂苷;鉴定了大鼠口服KXS-60%E后血中的5个原型皂苷类成分及4个代谢产物。结论:远志皂苷类成分及4个代谢产物可能是KXS-60%E潜在的活性成分及直接药效物质基础,这将有助于阐明开心散防治阿尔兹海默症的有效成分及作用机制。
OBJECTIVE: To identify the chemical components of Polygala tenuifolia and its metabolites of Polygala tenuifolia in 60% ethanol extract (KXS-60% E) and its blood components. METHODS: Secondary ion information from TOF-MS / MS was analyzed by UPLC-TOF-MS. Based on Mass Lynx v4.1 analysis software and existing literature databases, the KXS-60% E COSCO saponin components, gavage of KXS-60% E in serum of Polygalaceae saponins components and their metabolites. Results: 14 Triterpenoid saponins of KXS-60% E were identified, mainly triterpenoid saponins. Five prototype saponins and 4 metabolites in the blood of KXS-60% E were identified. CONCLUSION: Polygala tenuifolia and its four metabolites may be the potential active ingredient of KXS-60% E and the direct drug substance basis, which will be helpful to elucidate the effective ingredients and mechanism of Kaixin San for the prevention and treatment of Alzheimer’s disease.
