目的探讨核胞浆转运蛋白α-5(karyopherin alpha-5,KPNA5)在重症肌无力(myasthenia gravis,MG)患者胸腺组织中的表达情况。方法分别利用荧光定量聚合酶链反应(fluorescence quantitative polymerase chain reaction,FQ-PCR)和免疫组化染色方法,检测MG组20例患者与对照组10名受试者胸腺组织中KPNA 5 mRNA和蛋白分子的表达。结果MG组和对照组胸腺组织中,KPNA 5 mRNA的表达水平分别为(0.71±0.08)和(0.57±0.02),两者之间的差异有统计学意义(P<0.05);KPNA 5蛋白阳性细胞均数分别为(8.99±0.49)和(5.88±0.69),差异有统计学意义(P<0.05)。结论KPNA 5分子在MG胸腺组织中表达增高,可能参与了MG的发生,是MG相关基因。
Objective To investigate the expression of karyopherin alpha-5 (KPNA5) in thymic tissue of patients with myasthenia gravis (MG). Methods The levels of KPNA 5 mRNA and protein in thymus of 20 subjects in MG group and 10 subjects in control group were detected by fluorescence quantitative polymerase chain reaction (FQ-PCR) and immunohistochemical staining respectively expression. Results The expression levels of KPNA 5 mRNA in MG group and control group were (0.71 ± 0.08) and (0.57 ± 0.02) respectively, with statistical significance (P <0.05). The positive expression of KPNA 5 protein The average cell numbers were (8.99 ± 0.49) and (5.88 ± 0.69), respectively, with significant difference (P <0.05). Conclusion The expression of KPNA 5 in MG thymus tissue may be involved in the genesis of MG and is related to MG.
