高效薄层分析技术(HPTLC)是近十年来迅速发展起来的一种微量分析方法,它具有点样量少,分辨率高,检出限量低等优点,适用于微量成分检出与定量。我所七室研究成功并生产了用高效粘合剂的硅胶薄层预制板(简称预制板),这为研究药物在动物体内外的生物转化提供了条件。我们采用了该种预制板建立了大鼠血中炔雌三醇环戊醚(CEE_3)及其代谢产物的薄层扫描测定法,以期深入研究药物的代谢规律,为临床用药提供资料。实验方法一、预制板性能:薄板大小10×10cm,薄板厚度0.2mm,硅胶粒度10—20μM。薄板使用前在110℃烘箱活化1小时。
High performance thin layer analysis (HPTLC) is a kind of micro analysis method developed rapidly in recent ten years. It has the advantages of less spotting amount, high resolution and low detection limit, which is suitable for the detection and quantification of trace components. My research in Room 7 succeeded in producing a thin layer of prefabricated silica gel sheet (referred to as prefabricated panels) with high efficiency binder, which provided the conditions for studying the biotransformation of drugs in and out of animals. We used this kind of prefabricated plate to establish the thin layer scanning assay of rat ethinylestradiol cyclopentenyl ether (CEE 3) and its metabolites, in order to study the metabolism regularity of the drug in detail and provide information for clinical drug use. Experimental methods First, the performance of prefabricated panels: sheet size 10 × 10cm, sheet thickness 0.2mm, silica particle size 10-20μM. The sheet was oven-activated for 1 hour at 110 ° C prior to use.
