目的探讨环孢素A(CsA)和雷公藤单体T4(T4)的免疫抑制机制以及两药联合使用的效应,特别是小剂量CsA、T4联合获得较强免疫抑制效应的可能性。方法用3H-TdR掺入率法测定人系膜细胞(MsC)增殖,用逆转录-聚合酶链反应(RT-PCR)和ELISA法测定细胞IL-6mRNA表达及培养上清IL-6含量。结果CsA和T4均剂量依赖性抑制系膜细胞增殖及IL-6合成,两药联合产生明显的协同作用,小剂量CsA、T4联合与大剂量CsA作用相当。结论CsA和T4具有协同免疫抑制作用
Objective To investigate the immunosuppressive mechanism of cyclosporin A (CsA) and Tripterygium wilfordii T4 (T4) and the combined effect of two drugs, especially the combination of CsA and T4 at low dose. Methods Proliferation of human mesangial cells (MsC) was measured by incorporation of 3H-TdR. The expression of IL-6 mRNA and the content of IL-6 in culture supernatant were determined by reverse transcription polymerase chain reaction (RT-PCR) and ELISA. Results Both CsA and T4 dose-dependently inhibited the proliferation and IL-6 synthesis of mesangial cells. The synergistic effect of the two drugs was observed. The combination of CsA and T4 at low dose was equivalent to that of CsA at high dose. Conclusion CsA and T4 have a synergistic immunosuppressive effect
