为探讨血管紧张素Ⅱ特异性受体 1阻滞剂氯沙坦对血浆血管紧张素Ⅱ及胸主动脉内皮功能的影响 ,腹腔注射链脲佐菌素复制大鼠糖尿病模型 ,分别于第 4、8、16周处死动物 ,取胸主动脉环测定乙酰胆碱引起的内皮依赖性舒张反应 ,并测定血浆血管紧张素Ⅱ浓度。结果发现 ,糖尿病大鼠随病程进展 ,其胸主动脉环乙酰胆碱引起的内皮依赖性舒张反应渐进性下降 ,血浆中血管紧张素Ⅱ浓度升高。提示 ,氯沙坦可明显改善内皮依赖性舒张反应 ,但对血浆血管紧张素Ⅱ浓度无降低作用。
To investigate the effects of losartan, an angiotensin Ⅱ-specific receptor 1 blocker, on endothelial function in plasma and angiotensin Ⅱ in rats, diabetic rats were injected intraperitoneally with streptozotocin (STZ) The animals were sacrificed on the 8th and 16th weeks. The thoracic aortic rings were used to determine the endothelium-dependent relaxation response induced by acetylcholine and the plasma concentrations of angiotensin Ⅱ were measured. The results showed that, with the progression of diabetic rats, the thoracic aorta induced by acetylcholine gradually decreased endothelium-dependent vasodilatation, plasma concentrations of angiotensin Ⅱ increased. Tip, losartan can significantly improve endothelium-dependent vasodilatory response, but no effect on plasma angiotensin II concentration.
