目的探讨白英提取液对Hela细胞凋亡及野生型P53(wtP53)和Bcl-2蛋白表达的影响。方法采用MTT法检测细胞增殖抑制率,荧光显微镜检测癌细胞形态变化,TUNEL法检测凋亡率,二步法免疫组化检测P53和Bcl-2蛋白表达变化。结果白英提取液能够抑制Hela细胞增殖,且呈剂量依赖性;观察到凋亡细胞典型的形态特征;细胞凋亡率随白英提取液浓度增加而升高;P53蛋白表达显著上升(P<0.01),而Bcl-2蛋白表达明显下降(P<0.01)。结论白英提取液能促进Hela细胞凋亡,其分子机制可能与上调野生型P53蛋白表达和下调Bcl-2蛋白表达有关。
Objective To investigate the effects of Baiying extract on the apoptosis of Hela cells and the expression of wild type P53 (wtP53) and Bcl-2 proteins. Methods MTT method was used to detect the cell proliferation inhibition rate. The morphological changes of cancer cells were detected by fluorescence microscopy. The apoptosis rates were detected by TUNEL method. The expressions of P53 and Bcl-2 protein were detected by two-step immunohistochemistry. Results Baiying extract could inhibit the proliferation of Hela cells in a dose - dependent manner. The typical morphological features of apoptotic cells were observed. The apoptosis rate of Hela cells increased with the increase of the concentration of Baiying extract, and the expression of P53 protein increased significantly (P < 0.01), while the expression of Bcl-2 protein decreased significantly (P <0.01). Conclusion Baiying extract can promote the apoptosis of Hela cells. The molecular mechanism may be related to up-regulating the expression of wild-type P53 and down-regulating the expression of Bcl-2.
