目的研究两种内源性睡眠促进物质褪黑素和油酰胺联合应用后对大鼠催眠作用的变化。方法大鼠腹腔注射褪黑素(2.5,10mg·kg-1),油酰胺(25,50mg·kg-1)或联合应用褪黑素(2.5,10mg·kg-1)与油酰胺(25,50mg·kg-1)后,立即记录大鼠脑电和肌电变化。结果虽然褪黑素(2.5mg·kg-1)和油酰胺(25mg·kg-1)单独应用没有对大鼠脑电图产生明显影响,联合应用后显著缩短睡眠潜伏期(TSO)和觉醒时间(W),并且延长了慢波睡眠(SWS)和总睡眠时间(TS)。褪黑素(10mg·kg-1)和油酰胺(50mg·kg-1)单独具有明显的催眠作用,但联合应用后快波睡眠(PS)减少,其它睡眠指标未见变化。结论褪黑素和油酰胺联合应用后对大鼠催眠作用的变化较为复杂,其机制应进一步深入研究。
Objective To investigate the hypnotic effects of two endogenous sleep-promoting substances, melatonin and oleamide, on rats. Methods Rats were intraperitoneally injected with melatonin (2.5,10 mg · kg -1), oleamide (25,50 mg · kg -1) or melatonin (2.5,10 mg · kg -1) with oleamide (25, 50mg · kg-1), the changes of EEG and EMG of rats were recorded immediately. Results Although the single application of melatonin (2.5mg · kg-1) and oleamide (25mg · kg-1) had no significant effect on the electroencephalogram in rats, the combination of TSO and waking time W) and prolonged slow wave sleep (SWS) and total sleep time (TS). Melatonin (10mg · kg-1) and oleamide (50mg · kg-1) alone had obvious hypnotic effects. However, fast wave sleep (PS) decreased after combined application, and no changes were observed in other sleep parameters. Conclusion The combination of melatonin and oleamide has a more complex hypnotic effect on rats, and its mechanism should be further studied.
