目的制备绿原酸完全抗原,对完全抗原进行鉴定。方法通过碳二亚胺(EDC)法把绿原酸与载体蛋白牛血清白蛋白和卵蛋白进行偶联制备,采用紫外扫描法对合成抗原进行了鉴定;用绿原酸完全抗原免疫豚鼠,设生理盐水对照组,绿原酸标准溶液对照组。间接酶联免疫法测定免疫豚鼠抗血清的效价。结果完全抗原偶联成功,测定偶联比20,符合动物免疫的要求。ELISA法检测豚鼠抗血清效价,完全抗原组豚鼠抗血清稀释1∶128为阳性,大于空白复孔A值2倍。结论成功制备绿原酸完全抗原。
Objective To prepare the complete antigen of chlorogenic acid and identify the complete antigen. Methods The chlorogenic acid was conjugated with carrier protein bovine serum albumin and egg protein by EDC method. The synthetic antigen was identified by ultraviolet scanning. The guinea pigs were immunized with the complete antigen of chlorogenic acid. Saline control group, chlorogenic acid standard solution control group. Indirect ELISA for determination of antibody titer in guinea pigs. The results of the complete antigen coupling success, determination of coupling ratio of 20, in line with the requirements of animal immunization. Antibody titer of guinea pigs was detected by ELISA. The complete antigen group guinea pig antisera diluted 1: 128 was positive, which was more than 2 times of A value. Conclusion Chlorogenic acid complete antigen was successfully prepared.
