目的:通过观察滋阴明目丸对视网膜光损伤大鼠模型Bcl-2、p53表达的影响,探讨滋阴明目丸对视网膜光损伤大鼠模型的保护作用及治疗机制。方法:将60只Sprague-Dawley大鼠随机分为空白对照组、模型对照组、阳性对照组、滋阴明目丸低剂量组、滋阴明目丸中剂量组、滋阴明目丸高剂量组6组,每组10只。空白对照组不予处理,其余均建立视网膜光损伤大鼠模型,造模后第3天予以灌胃,连续15 d,其中滋阴明目丸低、中、高剂量组予以不同剂量滋阴明目丸灌胃,阳性对照组予以杞菊地黄丸灌胃。取大鼠眼球做免疫组化检测Bcl-2及p53的表达。结果:Bcl-2免疫组化结果:与空白对照组比较,模型对照组有统计学差异(P=0.025<0.05);与模型对照组比较,阳性对照组(P=0.003<0.01)、中剂量组(P=0.004<0.01)及高剂量组(P=0.001<0.01)均有显著统计学差异,低剂量组无明显差异(P>0.05)。p53免疫组化结果:与空白对照组比较,模型对照组有显著统计学意义(P=0.000<0.01);与模型对照组比较,低剂量组无明显差异(P>0.05);阳性对照组(P=0.012)、中剂量组(P=0.002)及高剂量组(P=0.002)均有统计学差异(P<0.05)。结论:视网膜光损伤模型可使大鼠视网膜组织抗凋亡蛋白Bcl-2表达降低,促凋亡蛋白p53表达增高,滋阴明目丸能有效增强Bcl-2的表达,降低p53的表达,从而对细胞凋亡起到双向调节作用,对视网膜光损伤模型具有一定的治疗作用。
OBJECTIVE: To observe the effect of Ziyin Mingmu Pill on the expression of Bcl-2 and p53 in retinal photodamaged rat model and explore the protective effect of Ziyin Mingmu Pill on retina photoinduced injury in rats and its therapeutic mechanism. Methods: 60 Sprague-Dawley rats were randomly divided into blank control group, model control group, positive control group, low dose of Ziyin Mingmu Pill, medium dose of Ziyin Mingmu Pill, high dose of Ziyin Mingmu Pill Group 6, each group of 10. The blank control group was not treated, the rest were established retinal photodamage rat model, the first 3 days after the model was intragastric administration, continuous 15 d, of which Ziyin Mingmu low, middle and high dose groups were given different doses of yin Ming Mouthwash gavage, positive control group to Qijudihuangwan intragastric administration. The eyeballs of rats were used for immunohistochemistry to detect the expression of Bcl-2 and p53. Results: Compared with the blank control group, the expression of Bcl-2 protein in the model control group was significantly different (P = 0.025 <0.05). Compared with the model control group, the positive control group (P = 0.003 < (P = 0.004 <0.01) and high dose group (P = 0.001 <0.01). There was no significant difference in low dose group (P> 0.05). Compared with the control group, there was no significant difference in the expression of p53 in the low-dose group (P> 0.05). Compared with the blank control group, the model control group had significant statistical significance (P = 0.000 <0.01) (P = 0.012), middle dose group (P = 0.002) and high dose group (P = 0.002) (P <0.05). Conclusion: Retinal light damage model can decrease the expression of anti-apoptotic protein Bcl-2 and increase the expression of pro-apoptotic protein p53 in retina of rats, and ZYCM can effectively enhance the expression of Bcl-2 and decrease the expression of p53 Play a two-way regulation of apoptosis on retinal light damage model has a certain therapeutic effect.
