建立了高效液相色谱法(HPLC)测定再造烟叶中柠檬黄含量的分析方法。采用ZORBAX SB-C18(5μm,4.6 mm×150 mm)色谱柱分离,以甲醇及20mmol/L乙酸铵水溶液为流动相梯度洗脱,测定波长为426 nm,外标法定量,在0.5~200μg/m L范围内,柠檬黄的峰面积与其质量浓度间呈良好的线性关系,相关系数(r2)为0.9998,检出限为0.086μg/m L,定量限为0.26μg/m L,在1、50、200μg/m L添加水平下的加标回收率为96.7%~99.2%,相对标准偏差RSD值小于5%。试验结果表明,该方法操作简单易行、精密度高、回收率和重现性好。
A high performance liquid chromatographic (HPLC) method for the determination of lemon yellow in reconstituted tobacco was developed. The separation was carried out on a ZORBAX SB-C18 column (5 μm, 4.6 mm × 150 mm) using a mobile phase of methanol and 20 mmol / L ammonium acetate as the mobile phase. The wavelength was 426 nm. The correlation coefficient (r2) was 0.9998, the detection limit was 0.086μg / mL, the limit of quantification was 0.26μg / mL, The spiked recoveries were 96.7% ~ 99.2% at 50 and 200 μg / mL, and the relative standard deviations (RSDs) were less than 5%. The experimental results show that this method is easy to operate, high precision, good recovery and reproducibility.
