目的评估显色原位杂交(CISH)技术在检测乳腺癌标本中 HER2基因扩增的应用价值。方法分别采用免疫组织化学(IHC)En Vision 和 CISH 两种方法,检测165例乳腺癌中 HER2蛋白表达及 HER2基因扩增的情况。结果 (1)IHC 检测 HER2蛋白表达阴性者107例,表达阳性1+者24例,CISH 均未检测到 HER2基因扩增;(2)IHC 检测 HER2表达3+者22例,CISH 检测中21例呈现 HER2基因的高倍扩增,仪1例呈低倍扩增,HER2基因高倍扩增及其蛋白表达之间的符合率为95.5%;(3)IHC 检测 HER2表达2+者12例,CISH 检测有3例 HER2基因呈高倍扩增,6例呈低倍扩增,3例无扩增。结论 CISH 在检测 HER2基因扩增与 IHC 检测的结果具有较高的一致性和敏感性,可以作为一种检测乳腺癌 HER2基因状况的方法。
Objective To evaluate the value of chromosomal in situ hybridization (CISH) in the detection of HER2 gene amplification in breast cancer specimens. Methods Immunohistochemistry (IHC) En Vision and CISH were used to detect HER2 protein expression and HER2 gene amplification in 165 cases of breast cancer. Results (1) HERC protein was detected by IHC in 107 cases, 24 cases were positive for 1+, and no amplification of HER2 gene was detected by CISH. (2) Twenty-two cases were detected by IHC in HER2 with 3+, 21 cases by CISH The results showed that the coincidence rate of HER2 gene amplification and its protein expression was 95.5%. (3) Twelve cases with HER2 expression in IHC were detected by CISH Three cases of HER2 gene showed high magnification, 6 cases showed low magnification, 3 cases without amplification. Conclusion The results of CISH detection of HER2 gene amplification and IHC have high consistency and sensitivity and can be used as a method to detect HER2 gene status in breast cancer.
