目的:评价益肾降压方对RPH大鼠的降压效果,对肾实质的保护作用,并研究其对肾脏病理组织形态的影响。方法:通过大鼠肾次全切除加高盐喂养法制备RPH动物模型,以卡托普利为对照评价该方不同剂量组的降压作用、剩余肾组织、超微结构的病理改变,并通过免疫组化检测cNOS、ET-1两种内皮由来因子的分布表达。结果:益肾降压方各剂量组降压作用与空白对照组比较均有显著性差异(P<0.05),可减轻肾脏病理改变,对肾脏局部ET-1表达呈抑制并对cNOS表达上调。结论:益肾降压方可以降低肾实质性高血压,减轻肾脏病理改变,其机制可能通过对肾脏局部ET-1表达、分泌的抑制和cNOS表达、分泌的上调两条途径来实现其降压、保护肾脏的作用。
Objective: To evaluate the antihypertensive effect of Yishen Jiangya Recipe on RPH rats, the protective effect on renal parenchyma, and to study its effect on renal histopathology. METHODS: A rat model of RPH was prepared by subtotal nephrectomy and high-salt feeding in rats. Captopril was used as a control to evaluate the antihypertensive effect, the remaining renal tissue, and the ultrastructural pathological changes of the different dose groups of the rats. Immunohistochemistry was used to detect the distribution and distribution of cNOS and ET-1. RESULTS: The antihypertensive effect of Yishen Jiangfang Decoction group had significant difference with the blank control group (P<0.05), which could reduce the renal pathological changes, inhibit the expression of local ET-1 in the kidney and increase the expression of cNOS. Conclusion: Yishen Jiangya Decoction can reduce renal parenchymal hypertension and reduce renal pathological changes. The mechanism may be achieved through two ways of inhibiting the expression and secretion of ET-1 in the kidney and up-regulating the expression and secretion of cNOS in the kidney. Protect the role of the kidneys.
