目的探讨大鼠蛛网膜下腔出血后脑水肿组织中水通道蛋白4(aquaproin 4,AQP4)的表达变化。方法采用自体血注入枕大池法建立蛛网膜下腔出血动物模型,原位杂交和免疫组化法的方法分别检测蛛网膜下腔出血后AQP4 mRNA和蛋白质表达变化,并用干湿重法测定脑含水量变化。结果蛛网膜下腔出血后8h,脑含水量和AQP4 mRNA及蛋白表达开始上升,2d达到高峰,1周后降低到正常水平;AQP4 mRNA及蛋白的表达和脑含水量呈显著正相关(r=0.9236~0.9509,P<0.01)。结论AQP4与蛛网膜下腔出血性脑水肿的形成密切相关。
Objective To investigate the expression of aquaporin 4 (AQP4) in brain edema after subarachnoid hemorrhage in rats. Methods The model of subarachnoid hemorrhage was established by autologous blood injection into occipital cistern and the expression of AQP4 mRNA and protein in subarachnoid hemorrhage were detected by in situ hybridization and immunohistochemistry. Water changes. Results 8h after subarachnoid hemorrhage, brain water content, AQP4 mRNA and protein expression began to rise, peaked at 2d, and then decreased to normal level one week later. There was a significant positive correlation between AQP4 mRNA and protein expression and brain water content (r = 0.9236 ~ 0.9509, P <0.01). Conclusion AQP4 is closely related to the formation of subarachnoid hemorrhagic brain edema.
