目的:筛选白术多糖脱色、脱蛋白最佳工艺。方法:以脱蛋白率与多糖损失率为指标,选择最佳脱蛋白方法;通过单因素和正交试验确定白术多糖最佳脱色条件。结果:三氯乙酸法(TCA)为最佳脱蛋白方法,白术多糖脱色的最佳条件是:温度60℃,时间15 m in,活性炭加入量为1%,pH值为5.5。结论:优化后的工艺条件稳定有效,可为白术多糖工业化生产提供理论依据。
Objective: To screen the best technology of decoloring and deproteinization of Atractylodes macrocephala polysaccharide. Methods: Taking the deproteinization rate and polysaccharide loss rate as indexes, the best deproteinization method was selected. The optimal decolorization conditions of Atractylodes macrocephala polysaccharides were determined by single factor and orthogonal experiment. Results: TCA was the best method for deproteinization. The optimal conditions for the decolorization of polysaccharide from Atractylodes macrocephala were as follows: the temperature was 60 ℃, the time was 15 min, the amount of activated carbon was 1% and the pH value was 5.5. Conclusion: The optimized process conditions are stable and effective, which can provide a theoretical basis for the industrialized production of polysaccharide from Atractylodes macrocephala.
