为了进一步研究苏丹Ⅰ、Ⅲ、Ⅳ对人胃癌细胞(SGC-7901)增殖的影响,采用流式细胞仪测定了DNA含量的变化,分析细胞周期的变化.结果表明:苏丹Ⅰ、Ⅲ、Ⅳ促进了SGC-7901细胞由G1期进入S期和G2期,表现为G1期细胞平均DNA含量下降,S期DNA含量上升,G2期DNA含量上升,细胞增殖指数(PI)增加,S期细胞指数(SPF)增加.其中苏丹Ⅲ在50μg.mL-1剂量时对SGC-7901细胞增殖影响非常显著(P<0.01),苏丹Ⅰ、Ⅳ在10μg.mL-1剂量时对SGC-7901细胞增殖影响非常显著(P<0.01).苏丹Ⅰ、Ⅲ、Ⅳ是通过影响细胞周期,从而调节SGC-7901细胞的增殖活动,促进细胞分化增殖.
In order to further study the effect of Sudan Ⅰ, Ⅲ and Ⅳ on the proliferation of human gastric cancer cell line SGC-7901, the change of DNA content and the change of cell cycle were detected by flow cytometry.The results showed that the promotion of Sudan Ⅰ, Ⅲ and Ⅳ SGC-7901 cells entered the S phase and G2 phase from G1 phase. The results showed that the average DNA content of cells in G1 phase decreased, the DNA content in S phase increased, the DNA content increased in G2 phase, the cell proliferation index (PI) increased and the S phase cell index SPF), the effect of Sudan Ⅲ on the proliferation of SGC-7901 cells was significant (P <0.01) at the dose of 50μg.mL-1, while the effect of SudanⅠandⅣon the proliferation of SGC-7901 cells at the dose of 10μg.mL-1 (P <0.01). Sudan Ⅰ, Ⅲ and Ⅳ regulate the cell cycle and regulate the proliferation of SGC-7901 cells and promote the proliferation and differentiation of cells.
