目的:建立纵条纹炭角菌菌丝体液体培养的最佳条件。方法:以菌丝体生物量为指标,采用单因素试验及正交优化试验,统计学方法确定纵条纹炭角菌液体培养的最佳条件。结果:单因素实验表明,碳源以麦芽糖、葡萄糖或玉米粉最佳,最佳氮源为黄豆饼粉,最佳碳氮比为5∶1,其他最佳培养参数为pH 6、接种2块菌饼、振荡培养7 d。K、Mg、P及VB1对菌丝体生长有明显的促进作用。正交试验表明,在麦芽糖4%、黄豆饼粉0.8%、pH 7、25℃及振荡培养13 d的条件下,菌丝体生物量最大。结论:液体培养优化条件下可大量获得纵条纹炭角菌菌丝体,菌丝体干重可达8.65 g/L以上。
Objective: To establish the optimum condition for liquid culture of mycelium of Nicotiana tabacum. Methods: Using mycelial biomass as an indicator, single factor test and orthogonal test were used to determine the optimum conditions for the liquid culture of C. albicans. Results: The single factor experiments showed that the best carbon source was maltose, glucose or corn flour, the best nitrogen source was soybean cake powder, the best carbon / nitrogen ratio was 5:1, the other best culture parameters were pH 6, inoculation 2 Bacterial cake, shaking culture 7 d. K, Mg, P and VB1 on mycelium significantly promote the growth. Orthogonal test showed that under the conditions of 4% maltose, 0.8% soybean meal, pH7,25 ℃ and shaking culture for 13 d, the biomass of mycelium was the largest. Conclusion: The mycelium of C. nosocomialis can be obtained in a large amount under the condition of liquid culture. The dry weight of mycelia is up to 8.65 g / L.
