Von H ippel-L indau(VHL)基因具有调节转录、稳定细胞生长相关基因和调节细胞周期的功能。VHL的基因产物pVHL是泛素连接酶的成分,有调节低氧诱导因子(HIF)转录因子的稳定性的作用。其抑癌作用为pVHL作用于特异性蛋白而发生破坏作用,主要是通过wt-pVHL对转录延长因子E longin复合物形成的抑制,从而发挥对细胞生长相关基因表达的调控作用,同时pVHL与细胞的增殖、凋亡和癌细胞的浸润、转移有关。VHL基因突变、缺失、重排和超甲基化与肾细胞癌(RCC)的发生密切相关。
