目的 神经胶质瘤是最常见的一种颅内原发恶性肿瘤,但目前发病机制尚不清楚.ruvB-like2是腺苷三磷酸酶类(AAA+)家族的成员,被认为可能在调控急性单核细胞性白血病细胞和肝癌细胞生长方面发挥重要作用.该研究首次探究ruvB-like2是否也是调控神经胶质瘤细胞生长的关键因子.方法 通过RNAi技术下调ruvB-like2在大鼠C6脑胶质瘤细胞中的表达,检测细胞增殖和凋亡情况,进一步通过流式细胞术和RT-PCR技术探究细胞凋亡的机制.结果 下调表达ruvB-like2后,C6脑胶质瘤细胞生长速度显著减慢(P<0.05),细胞发生强烈凋亡.进一步探究发现,细胞凋亡的机制涉及,将细胞周期阻滞在G2/M期以及诱导促凋亡蛋白的上调表达.结论 该研究证明ruvB-like2是调控脑胶质瘤细胞生长的关键分子.研究结果为阐明脑胶质瘤细胞的发病机制,以及以ruvB-like2开发新型治疗药物提供依据.“,”Objective Glioma is one of the most common intracranial primary malignant tumors.However,the mechanism of pathogenesis is still unknown.ruvB-like2,a member of the AAA+ family,may play a role in regulating the growth of acute monocytic leukemia cells and liver cancer cells.The study was the first to investigate whether ruvB-like2 is a key factor in regulating the growth of glioma cells.Methods RNAi was used to down-regulate the expression of ruvB-like2 in rat C6 glioma cells.Proliferation and apoptosis of cells were measured,and the mechanism of apoptosis was also investigated by flow cytometry and RT-PCR.Results After down-regulation of ruvB-like2,the growth rate of C6 glioma cells slowed down significantly (P<0.05) and apoptosis occurred.Further investigation revealed that the mechanism of apoptosis involves the arrest of cell cycle in the G2/M phase and the up-regulation of Pro-apoptotic proteins.Conclusion This study demonstrated that ruvB-like2 plays a role in regulating the growth of glioma cells.The results provide a basis for elucidating the pathogenesis of glioma and for developing new therapeutic agents by targeting ruvB-like2.
