目的研究经低浓度MNNG转化的GES-1细胞(MC细胞)中Shh、Gli-1mRNA及蛋白的表达,以探讨Shh信号通路与胃癌前病变的关系。方法低浓度MNNG(N-甲基-N’-硝基-N-亚硝基胍)诱导GES-1(人永生化胃黏膜上皮细胞)转化为MC细胞,对照组为GES-1细胞。用RT-PCR检测ShhmRNA与Gli-1mRNA的表达,免疫细胞化学法检测Shh与Gli-1蛋白的表达。结果 Shh、Gli-1mRNA及蛋白在GES-1细胞中无表达,在MC细胞中表达阳性。结论 Shh信号通路可能参与了胃癌前病变过程。
Objective To investigate the expression of Shh and Gli-1 mRNA and protein in GES-1 cells (MC cells) transformed with low concentration of MNNG in order to investigate the relationship between Shh signaling pathway and gastric precancerous lesions. Methods GES-1 (human immortalized gastric epithelial cells) was transformed into MC cells with low concentration of MNNG (N-methyl-N’-nitro-N-nitrosoguanidine), and the control group was GES-1 cells. The expression of Shhm and Gli-1 mRNA was detected by RT-PCR and the expression of Shh and Gli-1 protein by immunocytochemistry. Results Shh, Gli-1 mRNA and protein were not expressed in GES-1 cells and were positive in MC cells. Conclusion Shh signaling pathway may be involved in the process of gastric precancerous lesions.
