目的:对多花黄精活性多糖的提取分离方法进行试验,确定最佳的提取分离方法,并对提取分离后的粗多糖的含量进行测定。方法:采用超声辅助水浴提取,醇沉分离得到多花黄精粗多糖。以葡萄糖为标准品,在488nm波长处,采用苯酚-硫酸法对多糖的含量进行测定。结果:葡萄糖在20~100mg/L质量浓度范围内呈良好的线性关系(r>0.999),RSD为0.23%~1.51%,平均加样回收率为100.2%。测定结果表明,不同产地的多花黄精活性多糖含量存在较大的差异。结论:本实验所建立的方法简便快速,准确高,可用于多花黄精多糖提取分离及含量测定的分析研究。
OBJECTIVE: To test the method of extraction and separation of polysaccharides from Polysaccharides, determine the best extraction and separation methods, and determine the content of crude polysaccharides extracted and separated. Methods: Ultrasonic-assisted extraction with water bath and alcohol sedimentation to obtain polygonatum polysaccharides. Glucose as a standard, at 488nm wavelength, the use of phenol - sulfuric acid method for the determination of polysaccharide content. Results: There was a good linearity (r> 0.999) for glucose in the range of 20-100 mg / L with RSD of 0.23% ~ 1.51% and average recovery of 100.2%. The results show that there are significant differences in the content of Polysaccharides from different habitats. Conclusion: The method established in this study is simple, rapid and accurate. It can be used for the extraction and separation and determination of polysaccharides from Pistacia chinensis.
