目的:观察燥湿解毒、助脾散精中药ZPSJ方对ICV-STZ模型大鼠海马CA1区总tau蛋白表达及Ser422位点磷酸化的影响。方法:SPF级健康雄性sprague-dawley(SD)大鼠28只,通过双侧侧脑室注射STZ复制SAD模型,分为假手术组、模型组、多奈哌齐组和ZPSJ治疗组。结果:各组大鼠海马CA1区总tau(tau-5)的阳性表达均无显著性差异(P>0.05);模型组大鼠海马CA1区P-tau-Ser422阳性表达的神经元明显增多,与假手术组相比有显著性差异(P<0.01);经过ZPSJ复方治疗后,P-tau-Ser422的阳性表达明显减少,与模型组比较有显著性差异(P<0.05),多奈哌齐组与模型组比较,无显著性差异(P>0.05)。结论:ZPSJ复方能够减少tau蛋白在Ser422位点的过度磷酸化,从而可能起到防止SAD病理改变形成的作用。
OBJECTIVE: To observe the effect of Zishang Powder, a Chinese herbal medicine for dispersing spleen and detoxication, on the expression of total tau protein and Ser422 phosphorylation in hippocampal CA1 area of ICV-STZ model rats. Methods: Twenty-eight SPF healthy male Sprague-dawley rats were divided into sham-operated group, model group, donepezil group and ZPSJ-treated group by bilateral STZ injection of SAD model. Results: There was no significant difference in total tau (tau-5) expression in hippocampal CA1 region of rats in each group (P> 0.05); P-tau-Ser422 positive neurons in hippocampal CA1 area of model group were significantly increased, (P <0.01). The positive expression of P-tau-Ser422 in ZPSJ group was significantly lower than that in sham operation group (P <0.01), and there was significant difference between model group and Donepezil group The model group, no significant difference (P> 0.05). Conclusion: ZPSJ compound can reduce tau hyperphosphorylation at Ser422 site, which may play a role in preventing the formation of SAD pathological changes.
