[目的]探讨卵巢上皮癌中整合素αvβ6、基质金属蛋白酶-9表达水平及临床意义。[方法]临床资料完整的卵巢上皮癌冷冻组织46例及正常卵巢组织10例作为对照。采用反转录—聚合酶链反应(RT-PCR)方法和免疫组织化学法检测αvβ6、MMP-9表达水平。[结果]①卵巢上皮癌中整合素αvβ6mRNA表达强度(0.895±0.189)显著性高于正常卵巢组织(0)(P=0.000);MMP-9mRNA表达强度(0.360±0.126)显著性高于正常卵巢组织(0.031±0.02)(P<0.001)。②卵巢上皮癌中整合素αvβ6阳性表达率为100%,而10例正常卵巢组织均阴性表达(P<0.05)。整合素αvβ6高表达与临床分期、病情进展、肿瘤部位密切相关(P<0.05),与年龄、病理类型、组织学分级、残留灶大小无关(P>0.05)。卵巢上皮癌中MMP-9阳性表达率为92.7%,正常卵巢组织阳性表达率为80%(P=0.000)。MMP-9高表达与临床分期、病情进展密切相关(P<0.05),与年龄、病理类型、组织学分级、残留灶大小、肿瘤部位无关(P>0.05)。③卵巢上皮癌中MMP-9的表达与αvβ6的表达呈正相关(r=0.801,P=0.000)。[结论]上皮性卵巢癌组织中,整合素αvβ6与MMP-9表达均上调,其表达上调与卵巢上皮癌侵袭性和不良预后有关。
[Objective] To investigate the expression of integrin αvβ6 and matrix metalloproteinase-9 in epithelial ovarian cancer and its clinical significance. [Methods] 46 cases of ovarian epithelial carcinoma with complete clinical data and 10 cases of normal ovarian tissue as control. The expression of αvβ6, MMP-9 was detected by reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry. [Results] ① The expression of integrin αvβ6 mRNA in ovarian epithelial carcinoma (0.895 ± 0.189) was significantly higher than that in normal ovarian tissue (0) (P = 0.000). The expression of MMP-9 mRNA (0.360 ± 0.126) Tissue (0.031 ± 0.02) (P <0.001). ② The positive expression rate of integrin αvβ6 in ovarian epithelial carcinoma was 100%, while it was negative in 10 cases of normal ovarian tissue (P <0.05). The high expression of integrin αvβ6 was closely related to clinical stage, disease progression and tumor location (P <0.05), but not to the age, pathological type, histological grade, residual tumor size (P> 0.05). The positive rate of MMP-9 in ovarian epithelial carcinoma was 92.7% and that in normal ovarian tissue was 80% (P = 0.000). The high expression of MMP-9 was closely related to the clinical stage and progression (P <0.05), but not to the age, pathological type, histological grade, residual tumor size and tumor location (P> 0.05). ③ The expression of MMP-9 in ovarian epithelial carcinoma was positively correlated with the expression of αvβ6 (r = 0.801, P = 0.000). [Conclusion] The expression of integrin αvβ6 and MMP-9 are both up-regulated in epithelial ovarian cancer tissues. The up-regulation of integrin αvβ6 and MMP-9 is associated with the aggressive and poor prognosis of epithelial ovarian cancer.
