目的 观察胆固醇合成抑制剂洛伐他汀 (lovastatin ,LOV)对HL 60细胞caspase 3基因表达的影响 ,进一步探讨其抑制细胞增殖、诱导细胞凋亡的分子机制。方法 4、8、16μmol/LLOV处理HL 60细胞 1~ 4d后 ,MTT比色法及生长曲线测定检测细胞增殖效应 ,流式细胞仪测定细胞周期相的分布及凋亡率 ,DNA梯形带检测观察细胞的凋亡效应 ,RT PCR及Westernblot方法分别分析LOV对HL 60细胞caspase 3mRNA、caspase 3蛋白表达的影响。结果 LOV能显著抑制HL 60细胞增殖 ,使细胞周期阻滞于G0 /G1期 ,细胞凋亡率增加 ,有凋亡特征性的DNA梯形带出现 ;LOV能上调HL 60细胞cas pase 3蛋白表达 ,但对caspase 3mRNA表达无明显影响 ,该作用呈剂量 效应和时间依赖关系。结论 LOV对HL 60细胞有增殖抑制和诱导凋亡作用 ,LOV能上调HL 60细胞caspase 3蛋白表达 ,推测这可能是LOV抑制HL 60细胞增殖并诱导其凋亡的重要分子机制。
Objective To observe the effect of lovastatin (LOV), a cholesterol synthesis inhibitor, on the expression of caspase 3 gene in HL 60 cells, and further explore the molecular mechanisms that inhibit cell proliferation and induce apoptosis. Methods HL 60 cells were treated with 4, 8 and 16 μmol/L LL for 1 to 4 days. MTT colorimetric assay and growth curve were used to detect cell proliferation. Flow cytometry was used to determine the distribution of cell cycle and apoptosis rate. The apoptotic effect of cells, RT PCR and Western blot analysis were used to analyze the effect of LOV on the expression of caspase 3 mRNA and caspase 3 protein in HL 60 cells. Results LOV significantly inhibited the proliferation of HL 60 cells, arrested the cell cycle in G0/G1 phase, increased the rate of cell apoptosis, and showed a DNA trapezoidal band characteristic of apoptosis. LOV could up-regulate the expression of cas pase 3 protein in HL 60 cells. However, there was no significant effect on the expression of caspase 3 mRNA. The effect was dose- and time-dependent. Conclusion LOV can inhibit proliferation and induce apoptosis in HL 60 cells. LOV can upregulate the expression of caspase 3 in HL 60 cells. It is speculated that this may be an important molecular mechanism for LOV inhibiting the proliferation of HL 60 cells and inducing apoptosis.
