目的建立拟反相高效液相色谱法测定市售减肥茶中咖啡因的含量。方法以极性硅胶柱作为分离柱,以乙腈-水(5∶95,V/V)为流动相,流速为1.0 ml/min,检测波长为274 nm,进样量为10μl。结果咖啡因在该色谱体系中,峰形佳,洗脱时间短,且与基体中的复杂杂质能有效分离,可以实现咖啡因的快速测定。其浓度为0.2μg/ml~107.0μg/ml时,本法的线性回归方程为y=27 834x+1 307.4,r=0.999 9。方法的最低检出限(S/N=3)为0.2μg/ml,回收率为98.6%~102.6%,相对标准偏差为1.6%~2.8%。结论该法简单、快速,仅5 min就能完成测定工作;同时,该方法的准确性好,线性范围宽,线性关系良好,而且流动相中乙腈的比例只占5%,极大降低了有机溶剂的消耗和排放,减少了对环境的污染,是现阶段最经济环保的一种液相色谱测试技术。
Objective To establish a reversed-phase high performance liquid chromatography (HPLC) method for the determination of caffeine in commercially available slimming tea. Methods The mobile phase was acetonitrile - water (5:95, V / V). The flow rate was 1.0 ml / min and the detection wavelength was 274 nm. The injection volume was 10μl. Results Caffeine in the chromatographic system, good peak shape, elution time is short, and with the complex matrix of impurities can be effectively separated, enabling rapid determination of caffeine. The linear regression equation of this method is y = 27 834x + 1 307.4, r = 0.999 9 when its concentration is 0.2μg / ml ~ 107.0μg / ml. The method has the lowest detection limit (S / N = 3) of 0.2μg / ml, the recovery of 98.6% -102.6% and the relative standard deviation of 1.6% -2.8%. Conclusions The method is simple and rapid, and the determination can be completed in just 5 min. Meanwhile, the method has good accuracy, wide linear range and good linearity, and the proportion of acetonitrile in the mobile phase is only 5%, which greatly reduces the organic Solvent consumption and emissions, reducing the pollution of the environment, is the most economical and environmentally friendly liquid chromatography test technology at this stage.
