目的观察皂角提取物对急性脑缺血再灌注损伤的保护作用。方法将SD雄性大鼠随机分为假手术组、脑缺血再灌注损伤模型组、复方丹参组及皂角提取物大、中、小剂量组(150、75、38 mg.kg-1.d-1)。用药组术前连续给药3 d,然后用线栓法造模,假手术组仅分离右侧颈总动脉、颈外动脉和颈内动脉,然后缝合伤口,其余各组大鼠分别于术后2 h拔线实现脑血流再灌注,24 h后观察神经功能评分,比较各组大鼠血清中丙二醛(MDA)和脑组织匀浆中NO以及髓过氧化物酶(MPO)的含量。结果皂角提取物能有效改善大鼠脑缺血再灌注后的神经功能,降低NO、MDA、MPO的含量。结论皂角提取物对急性脑缺血再灌注损伤有保护作用。
Objective To observe the protective effect of saponin extract on acute cerebral ischemia reperfusion injury. Methods Male Sprague-Dawley rats were randomly divided into sham operation group, cerebral ischemia reperfusion injury model group, compound salvia miltiorrhiza group and saponin extract large, medium and low dose groups (150,75,38 mg.kg-1.d). -1). The drug group was administered continuously for 3 days before surgery, and then was modeled with a thread thrombectomy method. The sham operation group separated only the right common carotid artery, external carotid artery, and internal carotid artery, and then the wound was sutured. The rest of the rats were administered postoperatively. After 2 h of drawing, cerebral blood flow reperfusion was performed. The neurological function scores were observed 24 h later. The contents of NO and MPO in malondialdehyde (MDA) and brain tissue homogenates were compared in each group of rats. . Results The saponin extract could effectively improve the neurological function and reduce the contents of NO, MDA and MPO after cerebral ischemia-reperfusion in rats. Conclusion The saponin extract has protective effects on acute cerebral ischemia-reperfusion injury.
