目的研究柴油机尾气颗粒物(DEPs)对小鼠精子质量的影响。方法21~22g健康成年ICR雄鼠随机分为对照组(PBS)、高剂量染毒组、中剂量染毒组、低剂量染毒组,每组6只。每组雄鼠乙醚麻醉后,通过咽后壁吸入的方式分别吸入PBS、360.0、90.0和24.0μg DEPs,每周2次,连续染毒4周,于第6周处死,检测并比较小鼠体重、睾丸及附睾重量,脏器(睾丸、附睾)系数、精子密度、总活力,体外受精率等指标。结果组间睾丸重量、睾丸系数比较差异无统计学意义(P>0.05)。剂量为360.0、90.0μg DEPs的小鼠染毒组与PBS组、24.0μg DEPs小鼠染毒组比较,附睾重量、附睾系数有显著增加(P<0.05);而精子密度、总活力、受精率显著下降(P<0.05)。总活力和受精率之间呈正相关(P=0.000)。结论中、高剂量染毒(360.0、90.0μg DEPs/小鼠)可以降低健康成年雄性ICR小鼠精子质量,低剂量(24. 0μg DEPs/小鼠)则无显著影响。
Objective To study the effects of diesel engine exhaust particulate matter (DEPs) on sperm quality in mice. Methods 21 ~ 22g healthy adult ICR male rats were randomly divided into control group (PBS), high dose exposure group, middle dose exposure group and low dose exposure group, 6 rats in each group. Rats in each group were anesthetized with ether and inhaled into PBS, 360.0, 90.0 and 24.0 μg of DEPs through the posterior pharyngeal wall respectively. The rats were exposed to DEPs twice a week for 4 weeks and sacrificed on the 6th week. The body weight of the mice , Testicular and epididymis weight, organ (testis, epididymis) coefficient, sperm density, total vitality, in vitro fertilization rate and other indicators. Results There was no significant difference in testicular weight and testicular coefficient between groups (P> 0.05). Compared with the PBS group and 24.0 μg DEPs mice, the epididymal weight and epididymal coefficient increased significantly (P <0.05) in the mice exposed to DEPs doses of 360.0 and 90.0 μg, while the sperm density, total vitality, fertilization rate Significantly decreased (P <0.05). There was a positive correlation between total activity and fertilization rate (P = 0.000). Conclusions Medium and high dose exposure (360.0 and 90.0 μg DEPs / mouse) can reduce sperm quality in healthy male adult male ICR mice with no significant effect at low doses (24.0 μg DEPs / mouse).
