目的:研究在严重烫伤大鼠肠黏膜组织中Wnt5a动态表达变化及其意义。方法:选取48只SD大鼠,将大鼠背部浸入恒温水浴盆(92℃)中20s达Ⅲ度烫伤,制作烫伤创面,采用随机数字法将大鼠分为对照组、单纯烫伤组(烫伤组)、烫伤后胰岛素治疗组(治疗组)。伤后取肠组织观察大体变化,取肠道黏膜HE染色后,行组织形态学观察,分别于烫伤后6,12,24,48h四个时相点观察,取肠黏膜组织以免疫组织化学的方法,检测Wnt5a的表达情况。结果:在烫伤组和治疗组Wnt5a均有显著表达,对照组有少量表达(P<0.05),伤后6-12hWnt5a的表达持续升高,烫伤组于12h开始表达逐渐减少,24h后表达又重新增多,治疗组在48h表达达到最高峰,与烫伤组相比,治疗组Wnt5a的表达水平明显降低(P<0.05),病理形态学变化显示烫伤早期大鼠肠黏膜组织损伤明显,治疗组大鼠肠黏膜组织损伤较烫伤组明显减轻。结论:严重烫伤早期SD大鼠肠黏膜组织损伤明显,Wnt5a可能是反映烧伤脓毒症的重要标志。
Objective: To study the dynamic changes of Wnt5a expression in the intestinal mucosa of severely scalded rats and its significance. Methods: Forty-eight Sprague-Dawley rats were dipped into the third-degree scald for 20s in a constant temperature hot-water bath (92 ℃) for burn wounds. The rats were randomly divided into control group, scald group ), Post-scald insulin treatment group (treatment group). After injury, the intestinal tissues were observed for gross changes. The intestinal mucosa were stained with HE and observed for histomorphology. At 4, 12, 24 and 48 hours after scald, the intestinal mucosa were examined by immunohistochemistry Methods to detect Wnt5a expression. Results: Wnt5a was significantly expressed in scalded and untreated groups, with little expression in control group (P <0.05). The expression of Wnt5a increased gradually from 6 to 12 h after injury, and decreased gradually from 12 h after scald injury, and resumed after 24 h (P <0.05). Compared with the scald group, the expression of Wnt5a in the treatment group was significantly decreased (P <0.05), and the histopathological changes showed that the injury of intestinal mucosa in the early stage of scald injury was obvious. The rats in the treatment group Intestinal mucosal injury than scalded significantly reduced. Conclusion: The injury of intestinal mucosa in SD rats after severe scald is obvious. Wnt5a may be an important marker of burn sepsis.
