以萌发7 d的大豆子叶节切断及其切片为外植体,研究了萌发培养基、不同激素及其配比对大豆子叶节愈伤组织诱导与分化的影响。结果表明:当不添加BA时,在子叶节切断处几乎无愈伤组织的产生,只有添加BA时,才能产生愈伤组织。外植体切取方式、愈伤组织诱导培养基中BA和TDZ的浓度配比对愈伤组织诱导率产生很大影响。BA浓度为0.05~0.15 mg·L-1,TDZ浓度为0.04~0.12 mg·L-1时,子叶节切断外植体的愈伤组织诱导率较低,平均不到20%;BA浓度为0.15 mg·L-1和TDZ浓度为0.08 mg·L-1时,愈伤组织诱导率最高,达到50%;而以子叶节切片为外植体时,愈伤组织诱导率得到极大提高,平均达到85%以上,最高可达95%。将诱导的愈伤组织转到含有不同浓度BA的分化培养基上诱导不定芽发生,BA浓度为0.2 mg·L-1时,不定芽诱导率最高可达21.4%。
The seedlings of Soybean cotyledonary node were cut off and their sections were used as explants for 7 days germination. The effects of germination medium, different hormones and their ratio on the induction and differentiation of soybean cotyledonary node were studied. The results showed that when BA was not added, there was almost no callus formation at the cotyledon node, and callus could be produced only when BA was added. The way of explant extraction, the ratio of BA and TDZ in callus induction medium had a great influence on the callus induction rate. When the concentration of BA was 0.05-0.15 mg · L-1 and the concentration of TDZ was 0.04-0.12 mg · L-1, the callus induction rate of explant in cotyledonary node was lower than 20% on average; the concentration of BA was 0.15 The callus induction rate reached 50% when the mg · L-1 and TDZ concentrations were 0.08 mg · L-1, while the callus induction rate was greatly increased when the cotyledonary node slices were used as explants.
