目的:观察桂枝芍药知母汤对免疫性关节炎大鼠血清中TNF-α与关节滑膜组织Bc l-2表达的影响,并初步探讨其治疗机制。方法:以弗氏完全佐剂诱导产生免疫性关节炎模型,用不同剂量桂枝芍药知母汤(GSZ汤20.6、10.3 g/kg)给模型动物灌胃用药,观察用药后动物血清TNF-α水平与致炎对侧膝关节滑膜组织Bc l-2表达。结果:桂枝芍药知母汤用药4w后,可明显使关节指数减小(P<0.05或P<0.01);使模型大鼠血清TNF-α水平降低(P<0.01);免疫组化染色镜检显示,大鼠膝关节滑膜组织Bc l-2阳性细胞数减少、阳性细胞着色强度减弱(P<0.05或P<0.01),本实验条件下,GSZ汤20.6 g/kg剂量时的疗效与MTX相当。结论:桂枝芍药知母汤对免疫性关节炎大鼠有治疗作用,其机制可能与抑制TNF-α分泌,降低Bc l-2表达有关。
OBJECTIVE: To observe the effect of Guizhi Shaoyao Zhimu Decoction on the expression of TNF-α in serum and Bc l-2 in synovial tissue of rats with immunological arthritis, and to explore its therapeutic mechanism. METHODS: The model of immune arthritis was induced with Freund’s complete adjuvant, and the model animals were intragastrically administered with different doses of Guizhi Shaoyao Zhimu Decoction (GSZ soup 20.6, 10.3 g/kg). The serum TNF-α was observed after administration. Level and Expression of Bcl-2 in the Synovial Tissue of Contralateral Knee Joint of Inflammatory. RESULTS: Guizhi Shaoyao Zhimu Decoction could significantly decrease the joint index (P<0.05 or P<0.01) after 4 weeks of treatment, and decreased the level of serum TNF-α in model rats (P<0.01). Immunohistochemical staining Examination showed that the number of Bcl-2 positive cells in the synovial tissue of the knee of the rat decreased and the positive cell staining intensity weakened (P<0.05 or P<0.01). Under the experimental conditions, the efficacy of the GSZ soup at the dose of 20.6 g/kg was MTX is quite. Conclusion: Guizhi Shaoyao Zhimu Decoction has therapeutic effects on rats with immune-induced arthritis, and its mechanism may be related to inhibiting the secretion of TNF-α and decreasing the expression of Bcl-2.
