目的对分离自脚骨脆正常植物组织中的内生真菌Leptosphaeria sp.L1201的代谢产物及其生物活性进行研究。方法采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、高效液相色谱等技术对内生真菌Leptosphaeria sp.L1201的代谢产物进行分离,采用核磁共振、高分辨率质谱、X-射线单晶衍射等技术确证化合物的结构;采用一氧化氮(NO)生成抑制试验评价化合物的抗炎活性,并测定化合物对小鼠RAW264.7巨噬细胞的细胞毒性。结果从该株内生真菌培养物的乙酸乙酯提取物中获得2个新的氧杂蒽酮类化合物,并鉴定为leptosphaerins H(1)和I(2);化合物1和2能够抑制小鼠RAW264.7巨噬细胞中NO形成,IC50值分别为126.7μmol/L和72.6μmol/L。结论 Leptosphaerins H(1)和I(2)为新化合物,并在RAW264.7细胞中具有一定的抗炎活性。
OBJECTIVE To study the metabolites of Leptosphaeria sp.L1201, an endophytic fungus, isolated from normal tissues of foot bones and their biological activities. Methods The metabolites of endophytic fungus Leptosphaeria sp. L1201 were isolated by silica gel column chromatography, Sephadex LH-20 gel column chromatography and high performance liquid chromatography (HPLC), and their metabolites were separated by 1H NMR, high resolution mass spectrometry, X-ray single crystal diffraction And other techniques to confirm the structure of the compound; the anti-inflammatory activity of the compound was evaluated using a nitric oxide (NO) production inhibition assay and the cytotoxicity of the compound to mouse RAW 264.7 macrophages was determined. Results Two new xanthone compounds were obtained from the ethyl acetate extracts of this strain of endophytic fungi culture and identified as leptosphaerins H (1) and I (2); Compounds 1 and 2 were able to inhibit mice NO was formed in RAW264.7 macrophages with IC50 values of 126.7 μmol / L and 72.6 μmol / L, respectively. Conclusion Leptosphaerins H (1) and I (2) are new compounds and have some anti-inflammatory activity in RAW264.7 cells.
