目的:探讨以高速逆流色谱(HSCCC)法分离丁公藤中东莨菪内酯的方法。方法:用HSCCC法对丁公藤粗提物进行分离,分离条件:两相溶剂系统为正己烷-乙酸乙酯-甲醇-水(3:5:3.5:5,V/V/V/V),上相为固定相,下相为流动相,转速为850r/min,流速为2ml/min,水浴温度为25℃。结果:通过红外光谱、质谱和核磁共振分析,得到的产物是东莨菪内酯;经高效液相色谱测定,产物纯度为97.89%。结论:该法可对丁公藤中东莨菪内酯进行快速有效地分离。
Objective: To explore the method for the separation of scopoletin from Ding Gongtu by high speed countercurrent chromatography (HSCCC). Methods: The crude extracts of Dinggong were separated by HSCCC. The separation conditions were: n-hexane-ethyl acetate-methanol-water (3: 5: 3.5: 5, V / V / V / V) The phase was stationary phase, the lower phase was mobile phase, the rotation speed was 850r / min, the flow rate was 2ml / min and the temperature of water bath was 25 ℃. Results: The product was scopoletin by IR, MS and NMR. The purity of the product was 97.89% by high performance liquid chromatography. Conclusion: This method can be used for the rapid and efficient separation of alloxan.
