分别提取热休克前后的激活淋巴细胞及Jurkat细胞的总(?)mRNA,以之为模板通过逆转录法得到以α-~(32)P标记的第一链cDNA,再以cDNA为探针同abl,myc,myb,fos,ras五种癌基因及TcR-β,IL-2,IL-2R,TfR四种与细胞生长增殖密切相关的基因片段进行斑点杂交。结果表明热休克可使激活的淋巴细胞和Jurkat细胞中c-myc和TfR mRNA的表达明显降低,而IL-2和Ki-rat的表达增加。并对热休克与基因表达的关系进行了初步讨论。
After extraction of heat shock, activated lymphocytes and total (?) mRNA of Jurkat cells were extracted and used as a template to obtain a first-stranded cDNA tagged with α-(32)P by reverse transcription. The cDNA was used as a probe. Four oncogenes (abl, myc, myb, fos, ras) and four gene fragments closely related to cell growth and proliferation of TcR-β, IL-2, IL-2R and TfR were used for dot blotting. The results showed that heat shock significantly decreased the expression of c-myc and TfR mRNA in activated lymphocytes and Jurkat cells, while the expression of IL-2 and Ki-rat increased. The preliminary discussion of the relationship between heat shock and gene expression was carried out.
