目的 探讨p5 3和Bcl- 2基因在实验性急性心肌梗死 (AMI)心肌细胞凋亡中的作用。方法 用TUNEL法检测 48只兔AMI模型梗死区的心肌细胞凋亡 ,用以cDNA为内参标的逆转录 聚合酶链反应 (RT -PCR)的方法检测其心肌细胞内p5 3和Bcl- 2基因mRNA表达量 ,并探讨它们之间的相互关系。结果 TUNEL法发现兔梗死区心肌细胞凋亡千分率在冠脉结扎前( 0h)和结扎后 1h、 2h、 3h、 4h、 6h、 8h、 12h的改变如下 :0h组和 12h组 <1h组 <2h组和 8h组<3h组 <4h组 (P均 <0 0 5 ) ;p5 3基因mRNA表达量依次为 :冠脉结扎后 4h组 >结扎后 6h组>结扎后 3h组 >结扎后 2h组 >未结扎 ( 0h)组和结扎后 1h组、 8h组、 12h组 (P均 <0 0 5 ) ;且梗死区心肌细胞凋亡数与其内p5 3mRNA表达量对数值之间成明显的正相关性 (r为 0 930 0 ,P<0 0 0 1)。Bcl- 2基因mRNA表达量 :冠脉结扎后 3h组、 4h组和 6h组 <结扎后 2h组、 8h组 <结扎后 1h组和未结扎 ( 0h)组 <结扎后 12h组 (P均 <0 0 5 ) ;且梗死区心肌细胞凋亡数分别与其内Bcl- 2mRNA表达量的对数值成明显的负相关性 (r为 - 0 8732 ,P <0 0 0 1) ;p5 3和Bcl- 2基因mRNA表达量的对数值之间亦呈明显的负相关性 (r=- 0 96 49,P <0 0 0 1)。结论 急性心肌梗死时心肌细胞存在明显的凋?
Objective To investigate the role of p53 and Bcl-2 genes in cardiomyocyte apoptosis in experimental acute myocardial infarction (AMI). Methods TUNEL method was used to detect the apoptosis of cardiomyocytes in the infarct zone of 48 rabbits with AMI. The mRNA of p53 and Bcl-2 was detected by reverse transcription polymerase chain reaction (RT-PCR) with cDNA as an internal standard The amount of expression, and explore the relationship between them. Results TUNEL assay showed that the percentage of cardiomyocyte apoptosis in rabbit infarction zone was significantly lower than that before ischemia (0h) and 1h, 2h, 3h, 4h, 6h, 8h, 12h after ligation <2h group and 8h group <3h group <4h group (P <0.05). The expression of p5 3 gene mRNA in the order of 4h after coronary ligation> 6h after ligation> 3h after ligation> 2h after ligation (P <0.05). There was a significant positive correlation between the number of apoptotic cardiomyocytes and the expression of p53mRNA in infarction area Correlation (r = 0 930 0, P <0 0 0 1). The expression of Bcl-2 mRNA was detected at 3h, 4h and 6h after coronary ligation, 2h after ligation, 8h after ligation, 1h after ligation and 0h before ligation (P <0.01). The number of apoptotic cardiomyocytes in infarction area was negatively correlated with the logarithm of Bcl-2 mRNA expression (r = -0.7732, P <0.01) The logarithm of gene mRNA expression also showed a significant negative correlation (r = - 0 96 49, P <0 0 0 1). Conclusion Myocardial cells have obvious apoptosis during acute myocardial infarction.
