背景:含有大黄的复方中药消可宁能有效防治早期糖尿病肾病。目的:观察大黄酸对髙糖培养的SD大鼠肾小球系膜细胞凋亡的影响。方法:以20,40,80μmol/L浓度的大黄酸刺激髙糖培养的肾小球系膜细胞,苏木精-伊红染色观察凋亡细胞形态,DAPI荧光染色观察细胞核凋亡情况,流式细胞仪观察细胞凋亡率。结果与结论:苏木精-伊红染色及DAPI染色结果显示大黄酸可促进髙糖培养的肾小球系膜细胞的凋亡,且与浓度与时间成正相关。各组肾小球系膜细胞的细胞凋亡率差异有显著性意义(P<0.05),表明大黄酸对髙糖培养的肾小球系膜细胞凋亡产生影响且与浓度及时间成正相关。
Background: Chinese herbal medicine containing rhubarb can be used to effectively prevent and treat early diabetic nephropathy. AIM: To observe the effect of rhein on the apoptosis of glomerular mesangial cells cultured with maltose in SD rats. Methods: Glomerular mesangial cells cultured in the presence of rhein at 20, 40 and 80μmol / L were stimulated with hematoxylin and eosin (HE). The morphological changes of apoptotic cells were observed by DAPI staining. The flow cytometry Cytometer observed the rate of apoptosis. RESULTS AND CONCLUSION: The results of hematoxylin-eosin staining and DAPI staining showed that rhein can promote the apoptosis of glomerular mesangial cells cultured in xylose and positively correlated with the concentration and time. The apoptosis rate of glomerular mesangial cells in each group was significantly different (P <0.05), indicating that rhein affected the apoptosis of glomerular mesangial cells cultured with xanthic acid and had a positive correlation with the concentration and time.
