目的:观察益气温阳中药坎离颗粒对腹主动脉缩窄诱导的压力过载性舒张性心力衰竭(DHF)大鼠血流动力学的影响。方法:清洁级雄性Wistar大鼠70只,腹主动脉缩窄术后,随机分为假手术组、模型组、中药组、缬沙坦组和中药+缬沙坦联合干预组(简称联合干预组),每组14只。分别于术后1、4、12、32周监测外周血压,并于32周后观察各组大鼠的血流动力学指标、心肌质量和左室组织形态。结果:与假手术组比较,模型组大鼠造模后第1、4、12、32周历时性前肢血压均明显升高,SAP、LVSP、LVEDP、CMI和LVMI显著升高,差异有统计学意义(P<0.05,P<0.01)。与模型组比较,中药组、缬沙坦组、联合干预组术后第4、12、32周前肢血压显著降低,中药组大鼠SAP、LVEDP降低,缬沙坦组、联合干预组SAP、LVSP、LVEDP降低,联合干预组HR、LVMI降低,缬沙坦组的CMI和LVMI降低,差异有统计学意义(P<0.05,P<0.01)。与中药组比较,联合干预组术后第12、32周前肢血压显著降低,差异有统计学意义(P<0.05)。与缬沙坦组比较,中药组降压幅度小,差异有统计学意义(P<0.05),而联合干预组和缬沙坦组相似。与中药组和缬沙坦组比较,联合干预组的LVEDP更低(P<0.05)。结论:益气温阳中药坎离颗粒能显著降低腹主动脉缩窄大鼠的外周血压,改善压力负荷超载诱导的舒张性心力衰竭。
Objective: To observe the effects of Yiqi Wenyang Kelm on hemodynamics of abdominal aorta-induced pressure overload-induced diastolic heart failure (DHF) in rats. Methods: Seventy clean male Wistar rats were randomly divided into sham operation group, model group, traditional Chinese medicine group, valsartan group and traditional Chinese medicine + valsartan combined intervention group ), 14 in each group. Peripheral blood pressure was monitored at 1, 4, 12, and 32 weeks after operation. The hemodynamics, myocardial mass and left ventricular morphology of the rats in each group were observed after 32 weeks. Results: Compared with the sham-operation group, the time-dependent extremities BPH, SAP, LVSP, LVEDP, CMI and LVMI significantly increased at the 1st, 4th, 12th and 32nd weeks in the model group Significance (P <0.05, P <0.01). Compared with the model group, the blood pressure in the forelimb of the Chinese medicine group, the valsartan group and the combined intervention group was significantly decreased at the 4th, 12th and 32nd weeks after operation, while the SAP and LVEDP were decreased in the traditional Chinese medicine group. In the valsartan group and the combined intervention group, SAP and LVSP (P <0.05, P <0.01). The LVEDP and LVEDP levels in the combined intervention group were lower than those in the combined intervention group. The CMI and LVMI were decreased in the valsartan group (P <0.05, P <0.01). Compared with the traditional Chinese medicine group, the blood pressure of forelimb in the intervention group decreased significantly at the 12th and 32nd week (P <0.05). Compared with the valsartan group, the decrease amplitude of the traditional Chinese medicine group was small, the difference was statistically significant (P <0.05), while the combination intervention group and valsartan group similar. Compared with the traditional Chinese medicine group and the valsartan group, LVEDP in the combined intervention group was lower (P <0.05). CONCLUSION: Kanqi Granule of Yiqi Wenyang can significantly reduce the peripheral blood pressure of abdominal aorta constriction rats and improve the diastolic heart failure induced by pressure overload.
