目的探讨白细胞介素-17(IL-17)在系统性硬皮病(SSc)患者中的表达水平,及其与结缔组织生长因子(CTGF)的相关性。方法选择68例SSc患者和同期整形外科收治的30例非SSc患者,采用酶联免疫吸附试验(ELISA)法检测血浆IL-17水平,采用免疫组化SABC法检测皮肤CTGF含量。比较各组IL-17和CTGF表达水平的差异,及二者的相关性。结果与对照组比较,SSc患者IL-17、CTGF蛋白表达水平较对照组显著升高,且活动组IL-17、CTGF蛋白表达水平显著高于非活动组(P<0.05);弥漫型SSc组IL-17、CTGF蛋白表达水平较局限型SSc组明显增高(P<0.05);SSc患者IL-17、CTGF蛋白与改良Rodman总皮肤厚度评分(MRSS)均呈线性正相关(r=0.712,0.738,P<0.05);血浆IL-17与皮损组织CTGF蛋白亦呈线性正相关(r=0.680,P<0.05)。结论 IL-17在SSc患者外周血中表达增高,与CTGF具有较好相关性,共同参与了SSc的发病过程,阻断Th17细胞活化、分泌IL-17,有望为SSc的治疗提供新的靶点。
Objective To investigate the expression of interleukin-17 (IL-17) in patients with systemic scleroderma (SSc) and its relationship with connective tissue growth factor (CTGF). Methods Sixty-eight SSc patients and 30 non-SSc patients were enrolled in the same period of orthopedic surgery. Plasma IL-17 level was measured by enzyme-linked immunosorbent assay (ELISA) and immunohistochemical SABC method. The differences of IL-17 and CTGF expression levels in each group were compared between the two groups. Results Compared with the control group, the expression of IL-17 and CTGF protein in SSc patients was significantly higher than that in control group, and the levels of IL-17 and CTGF protein in active group were significantly higher than those in inactive group (P <0.05). The diffuse SSc group The expression of IL-17 and CTGF protein in SSc group was significantly higher than that in SSc group (P <0.05). There was a positive correlation between IL-17 and CTGF protein and modified Rodman’s total skin thickness score (MRSS) in patients with SSc (r = 0.712,0.738 , P <0.05). There was also a positive correlation between plasma IL-17 and CTGF protein in skin lesions (r = 0.680, P <0.05). Conclusion The expression of IL-17 in peripheral blood of patients with SSc is increased, which is correlated with CTGF. It participates in the pathogenesis of SSc, blocks the activation of Th17 cells and secretes IL-17, which may provide a new target for the treatment of SSc .
