目的:探讨14-3-3σ和cyclin B1蛋白在乳腺癌组织中的表达及其与乳腺癌临床病理特征之间的关系。方法:选择2006年11月至2009年11月在辽宁医学院附属第一医院手术治疗的乳腺癌患者95例乳腺癌和30例癌旁组织(距癌边缘5 cm)的标本,患者年龄为31~85岁,中位年龄58岁。免疫组织化学PV法和Western blotting检测14-3-3σ和cyclin B1蛋白在乳腺癌和癌旁组织中的表达,分析两者与乳腺癌临床病理特征之间的关系以及两者的相关性。结果:Western blotting与免疫组化实验结果显示,14-3-3σ蛋白在乳腺癌中表达率为22.1%(21/95),显著低于癌旁组织的93.3%(28/30)(P<0.01);14-3-3σ蛋白的表达与乳腺癌组织学分级、TNM分期、淋巴结转移有关(P<0.05)。Cyclin B1蛋白在乳腺癌中的表达率为69.5%(66/95),显著高于癌旁组织的40%(12/30)(P<0.05);CyclinB1蛋白的表达与乳腺癌淋巴结转移相关(P<0.05)。乳腺癌组织中14-3-3σ和cyclin B1蛋白的表达呈负相关(r=-0.333,P=0.001)。结论:14-3-3σ和cyclin B1蛋白与乳腺癌的发生、转移相关,两者联合检测可为乳腺癌诊断和治疗提供参考。
Objective: To investigate the expression of 14-3-3σ and cyclin B1 protein in breast cancer and its relationship with clinicopathological characteristics of breast cancer. Methods: From November 2006 to November 2009, 95 cases of breast cancer and 30 cases of paracancerous tissues (5 cm from the edge of cancer) were surgically treated in the First Affiliated Hospital of Liaoning Medical University. The patient’s age was 31 ~ 85 years old, the median age of 58 years old. The expression of 14-3-3σ and cyclin B1 protein in breast cancer and paracancerous tissues was detected by immunohistochemistry PV method and Western blotting. The relationship between them and clinicopathological features of breast cancer was analyzed. Results: The results of Western blotting and immunohistochemistry showed that the expression rate of 14-3-3σ protein in breast cancer was 22.1% (21/95), significantly lower than that in paracancerous tissues (93.3%, 28/30) (P < 0.01). The expression of 14-3-3σ protein was correlated with histological grade, TNM stage and lymph node metastasis (P <0.05). The expression of Cyclin B1 protein in breast cancer was 69.5% (66/95), which was significantly higher than that in paracancerous tissues (12/30) (P <0.05). The expression of Cyclin B1 was correlated with lymph node metastasis P <0.05). The expression of 14-3-3σ and cyclin B1 in breast cancer tissues was negatively correlated (r = -0.333, P = 0.001). CONCLUSION: The expressions of 14-3-3σ and cyclin B1 are correlated with the occurrence and metastasis of breast cancer. The combined detection of 14-3-3σ and cyclin B1 may provide reference for the diagnosis and treatment of breast cancer.
