作为活血化瘀传统中药,丹参具有改善红细胞的变形性的显著临床作用,其对红细胞膜直接作用目前尚不清楚。本研究采用显微共聚焦拉曼技术,测量丹参注射液作用前后人红细胞的拉曼光谱变化。结果显示丹参注射液作用后红细胞膜部分谱线对应的基团构象发生微小变化,归属于磷脂分子脂酰基C-C骨架反式构象的1 064,1 126cm-1谱线强度明显减弱,归属于磷脂分子脂酰基C-C骨架扭曲构象的1 091 cm-1谱线强度明显增强,磷脂链内纵向有序性参数(Strans)明显变小,提示红细胞胞膜在丹参注射液作用下胞膜磷脂分子脂酰基C-C骨架的反式构象转变为扭曲构象,增强红细胞膜磷脂流动性,增强细胞膜的流动性,丹参注射液对红细胞膜结构和功能的改善可为其发挥活血化瘀的中药药理作用的可能内在机制。
As a traditional Chinese medicine for promoting blood circulation and removing blood stasis, Salvia miltiorrhiza has a significant clinical effect of improving the deformability of erythrocytes and its direct effect on the erythrocyte membrane is not yet clear. In this study, microscopy confocal Raman technique was used to measure the Raman spectra of human erythrocytes before and after Danshen injection. The results showed that there was a slight change in the group conformation corresponding to some spectral lines of erythrocyte membrane after administration of Danshen injection. The intensity of 1 064,1 126 cm-1 line belonging to the transconjugation of lipid acyl CC skeleton was obviously weakened and was attributed to phospholipid molecules The 1 091 cm-1 spectral intensity of the distorted conformation of the acyl CC backbone was significantly increased, and the longitudinal order parameter (Strans) in the phospholipid chain was significantly reduced, suggesting that the membrane phospholipid fatty acyl CC The transconformation of the framework transforms into a twisted conformation, enhancing the fluidity of the erythrocyte membrane phospholipid and enhancing the fluidity of the cell membrane. The improvement of the structure and function of the erythrocyte membrane by the Danshen injection may be a possible intrinsic mechanism of its pharmacological effects.
