目的观察小檗碱对体外培养人前列腺癌PC3细胞凋亡的作用。方法体外培养前列腺癌PC3细胞,分别用10、20和40μmol/L小檗碱处理细胞后,采用MTT法检测其对细胞增殖的影响,20μmol/L小檗碱处理细胞后流式细胞术检测细胞周期,吖啶橙染色法检测细胞凋亡情况,并通过Western印迹法检测Bcl-xL及前列腺特异性抗原(PSA)蛋白表达。结果小檗碱处理后,PC3细胞出现增殖抑制,细胞周期停滞于G2/M期,吖啶橙染色呈黄染,PSA及Bcl-xL蛋白表达明显下调。结论小檗碱可诱导人前列腺癌PC3细胞凋亡,其作用机制可能与Bcl家族信号通路有关。
Objective To observe the effect of berberine on the apoptosis of human prostate cancer PC3 cells cultured in vitro. Methods Prostate cancer PC3 cells were cultured in vitro and treated with 10, 20 and 40 μmol / L berberine, respectively. The cell proliferation was detected by MTT assay. Cells were treated with 20 μmol / L berberine and analyzed by flow cytometry Cell cycle and Acridine orange staining were used to detect the cell apoptosis. Western blotting was used to detect the expression of Bcl-xL and prostate-specific antigen (PSA). Results After treated with berberine, the proliferation of PC3 cells was inhibited. The cell cycle arrest was at G2 / M phase. Acridine orange staining was yellow staining and the expressions of PSA and Bcl-xL protein were down-regulated. Conclusion Berberine can induce the apoptosis of human prostate cancer PC3 cells, and its mechanism may be related to the Bcl family signaling pathway.
