目的通过对比观察补肾填精方左归丸和左归丸去补阳药对去卵巢骨质疏松模型大鼠胶原代谢的影响,探讨左归丸阳中求阴法防治骨质疏松症的作用机制。方法取雌性SD大鼠32只,采用双侧卵巢摘除术建立骨质疏松模型,随机分为假手术组(Sham)、模型对照组(OVX)、左归丸组(ZGP)、左归丸去补阳药组(ZGPQ),每组8只。灌胃给药12周后收集各组大鼠血清和股骨组织,采用ELISA法检测血清中PICP和ICTP水平,采用Masson染色法检测股骨组织胶原纤维形态,采用免疫组织化学染色法检测股骨组织ColⅠ蛋白表达水平。结果与Sham比较,OVX血清PICP和ICTP水平显著升高(P<0.01)、股骨组织ColⅠ蛋白表达显著降低;与OVX比较,补肾填精方下调血清PICP和ICTP水平(P<0.05,P<0.01)、上调股骨组织ColⅠ蛋白表达。结论补肾填精方左归丸对去卵巢引起的骨质疏松症具有一定的防治作用,其作用机制可能与其干预胶原代谢有关,全方效果较为明显。
Objective To compare and observe the effect of Zuogui Pill and Zuo Gui Pill in promoting the metabolism of collagen in rats with ovariectomized osteoporosis by Zu Gui Wan and Zuo Gui Pian to explore the mechanism of Zuogui Wan Yangzhong for controlling osteoporosis. Methods Thirty-two female Sprague-Dawley rats were randomly divided into sham operation group, model control group (OVX), Zuguiwan group (ZGP), Zuoguiwan Yang composition group (ZGPQ), each group of eight. Serum and femur tissues of rats in each group were collected after intragastric administration for 12 weeks. Serum PICP and ICTP levels were detected by ELISA. Collagen fiber morphology of femur was detected by Masson staining. Col Ⅰ protein of femur was detected by immunohistochemical staining The expression level. Results Compared with sham group, the levels of PICP and ICTP in OVX serum were significantly increased (P <0.01) and the expression of Col Ⅰ protein in femur tissue was significantly decreased. Compared with OVX, Bushen Recipe down-regulated serum PICP and ICTP levels (P <0.05, P <0.01) ), Increased the expression of Col Ⅰ protein in femur. Conclusion Zuogui pill Prescription for ovariectomy-induced osteoporosis has a certain role in prevention and treatment, its mechanism may be related to its intervention in the metabolism of collagen, the full effect is more obvious.
