目的了解从人体抽吸的脂肪组织中获得的脂肪干细胞在体外向成脂和成软骨细胞诱导分化的情况。方法临床提取8例人脂肪抽吸组织中的间充质干细胞,在体外分别进行成脂和成软骨诱导分化,并通过油红O染色、阿尔辛蓝染色和免疫组化的方法进行证实。结果脂肪干细胞经成脂诱导后,向成脂细胞分化,细胞内出现油红O染色阳性脂滴;经成软骨诱导后,细胞向成软骨细胞分化,分泌软骨特异性基质成分硫酸蛋白聚糖和Ⅱ型胶原。结论在适当的诱导条件下,来源于人体多部位脂肪抽吸组织中的干细胞具有向成脂和成软骨细胞分化的能力,本实验为进一步应用人体脂肪干细胞进行脂肪和软骨组织工程研究打下实验基础。
OBJECTIVE: To understand the in vitro differentiation of adipose-derived stem cells derived from human aspirated adipose tissue into adipogenic and chondrogenic cells. METHODS: Mesenchymal stem cells (MSCs) from 8 human aspirate tissues were extracted clinically and were differentiated into adipocytes and chondrocytes in vitro. The results were confirmed by oil red O staining, Alcian blue staining and immunohistochemistry. Results After induced by adipogenic differentiation, adipose-derived stem cells differentiated into adipogenic cells, and lipid droplets were observed in the cells. After induced by chondrocytes, the cells differentiated into chondrogenic cells and secreted cadherin-specific matrix components of sulfatase and Type II collagen. Conclusion Stem cells derived from adipose tissue from multiple body parts of human body have the ability to differentiate into adipogenic and chondrogenic cells under appropriate inducing conditions. This experiment laid the experimental foundation for the further study of fat and cartilage tissue engineering using human adipose-derived stem cells .
