目的:研究茵芩清肝汤对酒精性肝病大鼠血清一氧化氮(NO)、谷胱甘肽过氧化物酶(GSH-Px)的影响。方法:按每天8~12 mL.kg-1ig 52度白酒(随时间延长而递增,第1周为8 mL.kg-1,第2周10 mL.kg-1,第3周起12 mL.kg-1,以后继续维持此量,直至实验结束)配合高脂肪饲料喂养,造成大鼠酒精性肝病模型。随机分为模型组、正常组、茵芩清肝汤(20 g.kg-1ig)组,硫普罗宁(5 mg.kg-1ig)组、水飞蓟宾(5 mg.kg-1ig)组进行比较。每组30只,分别于实验第4周、第8周、第12周每组各取大鼠10只,称重后以乙醚麻醉动物,迅速断头取血,按常规方法分离血清,测量NO,GSH-Px含量。结果:模型组大鼠血清NO各阶段水平较正常组(15.36±7.2)μmol.L-1明显升高(P<0.01),第12周达到(40.8±7.5)μmol.L-1(P<0.01),GSH-Px各阶段水平较正常组(146.3±51.9)U.mg-1明显降低(P<0.01),第12周达到(86.4±6.4)U.mg-1(P<0.01)。各治疗组大鼠血清NO各阶段水平较模型组均降低(P<0.05),茵芩清肝汤组第12周达到(31.12±4.7)μmol.L-1(P<0.01);GSH-Px水平(107.7±31.1)U.mg-1较模型组均明显升高(P<0.01)。结论:茵芩清肝汤可降低血清NO含量、提高GSH-Px活性,抑制脂质过氧化反应,这可能是它防治酒精性肝病的机制之一。
Objective: To study the effect of Yinqin Qinggan Decoction on serum nitric oxide (NO) and glutathione peroxidase (GSH-Px) in alcoholic liver disease rats. Methods: According to the daily dose of 8 ~ 12 mL.kg-1ig 52-degree liquor (increasing with time, 8 mL.kg-1 in the first week, 10 mL.kg-1 in the second week and 12 mL in the third week. kg-1, continue to maintain this amount until the end of the experiment) with high-fat feed, resulting in alcoholic liver disease model in rats. The rats were randomly divided into 4 groups: model group, normal group, Yin Qin Qing Gan Tang (20 g.kg-1ig), tiopronin (5 mg.kg-1ig), silybin (5 mg.kg-1ig) Compare. 30 rats in each group. Ten rats in each group were taken at the 4th, 8th and 12th week of the experiment respectively. Animals were weighed and anesthetized with diethyl ether. Blood samples were rapidly decapitated and the serum was separated by routine methods. NO , GSH-Px content. Results: The levels of serum NO in model group were significantly higher than those in normal group (15.36 ± 7.2) μmol.L-1 (P <0.01) and reached (40.8 ± 7.5) μmol.L-1 in the 12th week (P < 0.01). The levels of GSH-Px in each phase were significantly lower than those in the normal group (146.3 ± 51.9 U.mg-1, P <0.01) and reached 86.4 ± 6.4 U.mg-1 in the 12th week (P <0.01). The levels of serum NO in each treatment group were lower than those in model group (P <0.05), Yinqin Qinggan Decoction group was (31.12 ± 4.7) μmol.L-1 in the 12th week (P <0.01) The level of (107.7 ± 31.1) U.mg-1 was significantly higher than the model group (P <0.01). Conclusion: Qinqin Qinggan Decoction can reduce serum NO content, increase GSH-Px activity and inhibit lipid peroxidation, which may be one of the mechanisms of prevention and treatment of alcoholic liver disease.
