目的:观察胶原三股螺旋重复蛋白1(CTHRC1)在胰腺癌组织中的表达及其对胰腺癌细胞生物学行为的影响。方法:采用实时定量PCR(RT-PCR)方法检测40例胰腺患者癌组织与癌旁组织中CTHRC1的表达。胰腺癌Panc28细胞转染pcDNA3.1-CTHRC1或CTHRC1 siRNA后,以未转染的Panc28细胞为对照,采用克隆形成实验检测CTHRC1对胰腺癌Panc28细胞增殖,伤口愈合实验和Boyden小室检测细胞迁移和侵袭能力。结果:RT-PCR结果显示,胰腺癌组织CTHRC1 mRNA表达水平明显高于癌旁组织(P<0.05)。与对照组Panc28细胞比较,转染pcDNA3.1-CTHRC1上调CTHRC1后,Panc28细胞克隆形成数明显增加、伤口宽度百分比明显减少、侵袭的细胞数明显增多,而转染CTHRC1 siRNA下调CTHRC1表达后,Panc28细胞以上指标呈反向变化,差异均有统计学意义(均P<0.05)。结论:胰腺癌组织在CTHRC1表达升高,CTHRC1可能通过促进胰腺癌细胞的增殖、迁移和侵袭能力而参与胰腺癌的发生与发展。
Objective: To observe the expression of CTHRC1 in pancreatic carcinoma and its effect on the biological behavior of pancreatic cancer cells. Methods: The expression of CTHRC1 in 40 cases of pancreatic cancer tissues and adjacent normal tissues was detected by real-time quantitative PCR (RT-PCR). Panc28 pancreatic cancer cells were transfected with pcDNA3.1-CTHRC1 or CTHRC1 siRNA, the untransfected Panc28 cells as a control, clonogenic assay was used to detect CTHRC1 Panc28 pancreatic cancer cell proliferation, wound healing and detection of Boyden cell migration and invasion ability. Results: RT-PCR results showed that the expression of CTHRC1 mRNA in pancreatic cancer tissues was significantly higher than that in adjacent tissues (P <0.05). Compared with the control group, Panc28 cells transfected with pcDNA3.1-CTHRC1 CTHRC1, Panc28 cell clone formation increased significantly, the percentage of wound width was significantly reduced, the number of invasive cells was significantly increased, while CTHRC1 siRNA transfected CTHRC1 downregulation, Panc28 The above indexes of cells were changed in reverse, the differences were statistically significant (P <0.05). Conclusion: The expression of CTHRC1 in pancreatic cancer tissue is elevated. CTHRC1 may be involved in the occurrence and development of pancreatic cancer by promoting the proliferation, migration and invasion of pancreatic cancer cells.
