Objective:To study the effects of thermal injury and twohit,induced by endotoxin,on the expression of tumor necrosis factor receptor 1(TNFR 1),and TNFR 2 in hepatic cells.To test the hypothesis that rhubarb can inhibit the expression of TNFR 1 and TNFR 2 in systemic inflammatory response induced by thermal injury and the twohit.Methods:Prospective and randomized trial.The rat burned model was used.Endotoxin at a dose of 20 mg/kg was continuously infused to animals 12 hours after burns and kept for 12 hours.The expression of TNFR 1,TNFR 2,in liver and the concentrations of TNFα,interleukin6(IL6) in plasma and hepatic tissues were observed.Results:Hybridization in situ revealed that thermal injury and the twohit induced by endotoxin could obviously increase the expression of TNFR 1 and TNFR 2 in hepatic cell surface.However,the TNFR 1 is prevailing.The rhubarb could downregulate the expression of both of the receptors.There was very high concentrations of TNFα in plasma and hepatic tissues after thermal injury and the twohit.Rhubarb evidently lowered the concentrations of TNFα in both plasma and hepatic tissues( P <0 01).It also decreased the concentrations of IL6 in plasma ( P <0 01).Conclusions:Rhubarb could downregulate the expression of TNFR 1 and TNFR 2 in hepatic cell surface after thermal injury and the twohit.It also decreased the concentration of TNFα and IL6 in plasma and hepatic tissues.
